Research into the biology of cancer often uses animal models to investigate molecular mechanisms of carcinogenesis. However, a major limitation of rodent models is that they are larger than humans and thus require larger sample sizes and more experiments. A guinea pig model of primary mammary cancer is a promising alternative because it is a much smaller species and can be easily maintained. Although the transformation of the initiation of tumor growth of guinea pig mammary epithelial cells in culture has been reported previously, the transformation of cells in vivo with this system has been difficult and the survival of the resulting mammary tumor-bearing guinea pig has not been reported. To address these limitations, we have developed a technique to induce guinea pig mammary carcinoma by injection of the MMTV-myc oncogene into the mammary glands of guinea pigs, followed by the development of mammary tumor nodules that can be transplanted into the flank region of recipient guinea pigs. The development of the mammary tumor is monitored by visual inspection, palpation, and bioluminescent imaging. A total of 34 animals were injected with MMTV-myc oncogene and each transplanted into the flank region with four tumor nodules. Animals that developed tumors were sacrificed and the mammary tumor nodules were harvested. The transplanted tumor was minced and cell suspensions were prepared using a standard method. Tumor cells (1 x 106) were transplanted into the fourth mammary fat pads of a recipient guinea pig. The development of the MMTV-myc-induced guinea pig mammary tumors and survival of the tumor-bearing animal has been well characterized. The breast tumor-bearing guinea pig is a unique model that can be used for the in vivo functional studies of tumor biology such as drug delivery, gene therapy, and drug development. This model also provides an alternative for evaluation of human breast cancer cell lines in animal models, compared to the xenograft models that are commonly used in oncology.
